No. That would go before launching DOSBox.
Seriously. Read the readme file. It won't hurt you. After that, if you are still clueless, check the guides section on the forum or even ask your questions here.

Have you read the readme file, Luke?
HINT: You need to mount some drives first (at least a hard-disk and a cd-rom drive if needed).
